Utilizing a combination of microdroplet generation, automated flow injection, and online MS analysis, the method proved effective for protein reduction/digestion, including complete interchain disulfide bond reduction and partial deglycosylation, with significant implications for understanding protein therapeutic candidates' structure and drug-antibody ratios (DAR) in Antibody Drug Conjugates (ADCs).
