In this session, I will go through the formulation development steps followed in R&D team at Labcorp for the preparation, optimization and characterization of nano-formulation. Case studies on the preparation of lipid nanoparticles for RNA encapsulation and animal testing will be provided.

Formulation development was able to co-encapsulate self amplifying RNA (MARV) (20 μg/mL) with 200 μg/mL of Toll-like receptor agonists.
The non-viral vectors (lipid nanoparticles) that produced for loading saRNA (MARV) and TLR adjuvants have suitable properties for immunological testing in animals.
The study shows that the addition of TLR agonists (adjuvants) can modify the immune response induced by the saRNA and changing the adjuvant type also affects the immune response.
